MicroRNAs were isolated from tissue samples and analyzed using reverse transcription polymerase chain reaction.</p><p><bold>Results.</bold><bold> </bold>After studying the Cancer Genome Atlas, microRNA-196a-5p and microRNA-200b-3p with differential expression in tissue samples of different histological types of TGCT were identified. These microRNA are significantly overexpressed in non-seminomas, and their expression levels significantly differ from expression levels in normal tissues and seminomas. The level of microRNA-196b-5p expression in teratomas obtained after retroperitoneal lymph node dissection is significantly higher than in seminomas, and microRNA-200b-3p expression level is significantly higher than in tissues with necrotic changes. ROC analysis has shown 89 % sensitivity and 79 % specificity for microRNA-196b-5p as a diagnostic marker. For microRNA-200b-3p, sensitivity and specificity were 75 and 89 %, respectively.</p><p><bold>Conclusion. </bold>In this study, we have shown for the first time that microRNA-196a-5p and microRNA-200b-3p have high diagnostic potential as markers for identification of various types of TGCTs including teratoma.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение. </bold>На сегодняшний день отсутствуют точные модели для прогнозирования морфологического строения герминогенных опухолей яичек (ГОЯ) до оперативного лечения. По данным литературы, в качестве нового диагностического маркера ГОЯ, а также для дифференциальной диагностики основных гистологических форм после проведенной химиотерапии возможно использование микроРНК.</p><p><bold>Цель</bold><bold> </bold><bold>исследования</bold><bold> </bold>– поиск новых микроРНК как маркеров для дифференциальной диагностики тератомы и живой опухолевой ткани.</p><p><bold>Материалы</bold><bold> </bold><bold>и методы. </bold>В исследовании были проанализированы свежезамороженные образцы тканей, полученные после орхифуникулэктомии (<italic>n</italic><italic> </italic>= 46) и забрюшинной лимфаденэктомии (<italic>n</italic><italic> </italic>= 42) у пациентов с диагнозом ГОЯ. Из образцов тканей выделяли микроРНК, которые анализировали с помощью полимеразной цепной реакции с обратной транскрипцией.</p><p><bold>Резуль</bold><bold>таты.</bold><bold> </bold>В результате изучения материалов Атласа ракового генома выбраны микроРНК-196a-5p и микроРНК-200b-3p с дифференциальной экспрессией в образцах опухолевой ткани различных гистотипов ГОЯ. Данные микроРНК достоверно гиперэкспрессируются в несеминомах, при этом уровни их экспрессии в этих ГОЯ значимо различаются с таковыми как в нормальных тканях, так и в семиномах. Уровень экспрессии микроРНК-196b-5p в тератомах, образцы которых получены после забрюшинной лимфаденэктомии, достоверно выше, чем в семиномах, а уровень экспрессии микроРНК-200b-3p достоверно выше, чем в ткани с некротическими изменениями после терапии. ROC-анализ показал чувствительность 89 % и специфичность 79 % при использовании в качестве диагностического маркера микроРНК-196b-5p.
