The obtained cells were cultured in Matrigel with different culture media for selection of the optimal one. Cell viability and growth rates were assessed using the MTS assay.</p><p><bold>Results.</bold> In this study, we successfully obtained two organoid cultures of prostate cancer cells and identified the most effective composition of culture medium. Using a cytotoxic test, it was shown that the obtained organoid cultures of prostate cancer cells had different sensitivity to docetaxel which was reflected in different inhibition of the tumor cell growth rate.</p><p><bold>Conclusion.</bold> The utilization of prostate cancer organoids to determine the best treatment approach is a highly promising experimental technology. Nevertheless, additional research is required before integration of this technology into clinical practice.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение.</bold> Исследование чувствительности полученных от пациентов опухолевых органоидов к противоопухолевым препаратам представляет собой перспективный подход к персонализированному выбору схемы лечения.</p><p><bold>Цель исследования </bold>– получение 2 культур опухолевых органоидов предстательной железы, подбор оптимального состава культуральной среды и оценка на полученных органоидных культурах эффективности химиотерапевтического препарата доцетаксел, применяемого для лечения рака предстательной железы.</p><p><bold>Материалы и методы.</bold> Исходную ткань диссоциировали с использованием гомогенизатора gentleMACS Octo. Полученные клетки культивировали в матриксе Matrigel с различными составами питательной среды для определения оптимального варианта. С помощью MTS-теста проводили оценку жизнеспособности и определяли скорость роста клеток.</p><p><bold>Результаты.</bold> В работе удалось успешно получить 2 органоидные культуры опухолевых клеток рака предстательной железы и подобрать оптимальный состав питательной среды для культивирования. С помощью цитотоксического теста было показано, что полученные органоидные культуры рака предстательной железы имели разную чувствительность к доцетакселу, что выражалось в разном ингибировании скорости роста опухолевых клеток в 2 линиях.</p><p><bold>Заключение.</bold> Использование полученных от пациентов опухолевых органоидов рака предстательной железы представляет собой перспективный подход к определению наилучшей схемы лечения пациента.
