Next, the cells were cultured in matrix Matrigel with addition of a serum-free complete nutrient medium. For histological analysis, organoids were fixed in a 10 % formalin solution, followed by staining with hematoxylin and eosin according to the standard protocol. Cell viability was assessed using MTS assay.<bold>Results. </bold>In this work, we generated a new culture of prostate cancer cells. The histological analysis confirmed that the resulting organoids consist of tumor epithelial cells. As a result of the cytotoxic test, it was shown that in this case docetaxel (82.9 %; <italic>p </italic>= 0.32) didn’t reduce statistically significantly the viability of prostate cancer cells compared to the control.<bold>Conclusion. </bold>The use of tumor organoids of prostate cancer for selection of an optimal treatment regimen is a promising experimental technology, however, further research is necessary for its introduction into practice.</p></abstract><trans-abstract xml:lang="ru"><p><bold>Введение. </bold>Перспективным экспериментальным подходом к персонифицированному выбору схем лечения является исследование чувствительности опухолевых клеток к лекарственным препаратам на <italic>in vitro </italic>моделях опухолевых органоидов.<bold>Цель исследования </bold>– получение культуры опухолевых органоидов предстательной железы и оценка эффективности на данной культуре химиотерапевтического препарата доцетаксел, применяемого для лечения рака предстательной железы.<bold>Материалы и методы. </bold>Исходную ткань диссоциировали с помощью гомогенизатора gentleMACS Octo. Далее клетки культивировали в матриксе Matrigel с добавлением бессывороточной полной питательной среды. Для проведения гистологического анализа органоиды фиксировали в 10 % растворе формалина с последующей окраской гематоксилином и эозином по стандартному протоколу. Оценку жизнеспособности клеток проводили с помощью MTS-теста.<bold>Результаты. </bold>В работе удалось успешно получить культуру опухолевых клеток рака предстательной железы. Гистологический анализ подтвердил, что полученные органоиды состоят из опухолевых эпителиальных клеток.
