In 2020, olaparib was approved in the Russian Federation as a monotherapy for metastatic castration-resistant prostate cancer with germline or somatic mutations of genes involved in DNA repair by homologous recombination, after progression on therapy with new hormonal drugs. We describe a review of the main olaparib clinical trials assessing its efficacy, safety and tolerance in metastatic castration-resistant prostate cancer patients.</p></abstract><trans-abstract xml:lang="ru"><p>В представленном обзоре литературы рассмотрена проблема лечения метастатического кастрационно-резистентного рака предстательной железы и его связь с соматическими и герминальными мутациями генов, отвечающих за репарацию путем гомологичной рекомбинации (homologousrecombinationrepair, HRR). В августе 2020 г. олапариб был одобрен в России в качестве монотерапии метастатического кастрационно-резистентного рака предстательной железы с герминальными или соматическими мутациями генов, участвующих в репарации ДНК путем гомологичной рекомбинации, после прогрессирования на фоне терапии новыми гормональными препаратами.
